Abstract (de) | Diese Arbeit setzt sich mit hochauflösenden Mikroskopieverfahren auseinander, insbesondere mit Strukturierter Beleuchtung (SIM) und Lokalisationsmikroskopie (SMLM). Dazu werden zunächst die grundlegenden Eigenschaften von Licht und dessen Wirken auf/in Materie vorgestellt, um die Funktionsweise der Fluoreszenzmikroskopie zu motivieren. Eine Einführung in Abbildungstheorie und Fourieroptik dient dem Verständnis der Auflösungsbegrenzung der Weitfeldmikroskopie. |
Abstract (en) | This thesis treats the topic of superresolution microscopy with a focus on structured illumination microscopy (SIM) and single molecule localisation microscopy (SMLM). In the beginning, the nature of light and common phenomena concerning its propagation and interaction with microscopic and macroscopic media are introduced in order to build the groundwork for fluorescence microscopy. The physics of imaging and Fourier optics are discussed to understand the intrinsically limited resolution of widefield microscopy. The techniques applied are dealt with theoretically and their realization in the Combo setup is explained. SIM and SMLM are installed in this microscope with the possibility to switch between the two modes. Up to five lasers are directly accessible, which allows multi-color experiments. The effective laser power output has been measured and the intensities available in the object plane in both microscope modes have been calculated. Finally, measurements on two types of biological samples have been performed. Short overviews on the biological background are given and problems discussed, as well as additional ideas to compensate them. |
