In Krebszellen spielt der ErbB2-Rezeptor eine wichtige Rolle für den Erhalt des Zellzyklus und der Proliferation der Zelle. Somit ist dieser Rezeptor ein entscheidendes Ziel bei der Bekämpfung von Tumoren. Für die Bekämpfung von Krebs stehen verschiedene Therapieverfahren, wie Chemotherapie, Bestrahlung und Antikörpertherapie zur Verfügung.
In der Zellmembran bendet sich auch das Protein Connexin 43. Dieses bildet die für die Zellen wichtigen Gap Junctions, die sich in der Zellmembran zum Austausch von Ionen und elektrischem Potential benden. Nach Strahlungseinwirkung und einigen Antikörpertherapien treten bei Patienten Anzeichen von Herzrythmusstörungen auf.
Aus diesen Gründen ist es wichtig, die Auswirkungen der verschiedenen Therapien auf den ErbB2-Rezeptor und das Protein Connexin 43 zu untersuchen.
Dazu werden die Konformationen der ErbB2- und Connexin 43-Proteine in der Brustkrebs-Zelllinie MCF7 nach den Behandlungen mit dem Antikörper Herceptin, dem Liganden Neuregulin 1 und Bestrahlung mittels hochauösender Lichtmikroskopie analysiert. Um bessere Aussagen über die Wirkung der Behandlungsarten treen zu können, wurde eine Auswertung der Abstände zwischen den Proteinen in den Bereichen Zellgrenze (direkter Zell-Zell-Kontakt), reine Zellperipherie und gesamte Zellperipherie (Zellgrenze und Zellperipherie) durchgeführt. Als Ergebnis dieser Arbeit ergab sich, dass jede einzelne Behandlungsart zu Veränderungen der Proteinanordnungen in der Zelle führt

Within cancer cells the ErbB2 receptor plays a major role regarding the maintenance of the cell cycle and cell proliferation. Consequently, this receptor is a crucial target in battle against tumours. Various treatment methods, such as chemotherapy, irradiation and antibody therapy are available in cancer therapies.
The protein Connexin 43 also is located within the outer cell membranes. It forms gap junctions important for the cells, which reside in the cell membrane for exchange of ions and electrical potential. After exposure to radiation and antibody therapies indications for induced cardiac arrhythmias appear in patients.
For these reasons it is important to examine the eects of the dierent therapies on the ErbB2 receptor respectively the protein Connexin 43.
In order to do this, the conformations of the ErbB2 and Connexin 43 proteins within the breast cancer cell line MCF7 are analysed via high resolution light microscopy after treatment with the antibody Herceptin, the ligand Neuregulin 1 and irradiation. To be able to make better statements about the impact of the types of treatment diverse evaluations were performed. These include the distances between the proteins in distinct areas, such as the cell border (direct cell to cell contact), the mere cell periphery as well as the entire cell periphery (cell border and cell periphery). As a result of this thesis it emerged, that every single type of treatment leads to alterations of the arrangements of the proteins in the cell.
