Abstract (en) | In this thesis the radiation dependence of the structure of heterochromatin and γ-H2AX, which is a protein that gathers around double-strand brakes and forms so called Foci, in nuclei of the SkBr3 cell line irradiated with 0Gy to 8Gy was investigated. To do that, a localisation microscope, which is able to take pictures with a resolution of about 10nm (which is much better than standard light micrsocopes are able to), was used to dtermine various parameters of γ-H2AX Foci and the density of heterochromatin in and around them. Based on a protocoll already radiated cells were marked with fluorecent dyes, targeting the histone proteins γ-H2AX and H3k9me3. After pictures were taken at the microscope, they were evaluated by different algorithms, which some of them were selfmade and others were developed by the research group around Prof. Dr. Michael Hausmann. |
